Helicase functions at the origin of replication to separate the two strands of the DNA double helix, forming the replication fork and single-stranded DNA templates for DNA polymerase. It is ATP-dependent, translocating along the DNA backbone and disrupting hydrogen bonds between complementary bases. In eukaryotes, the MCM (minichromosome maintenance) helicase complex, part of the pre-replication complex, performs this function, loaded onto DNA by ORC (origin recognition complex) during G1 phase and activated at the G1/S transition via CDK and DDK kinase activity. In prokaryotes, DnaB helicase serves this role, loaded by DnaC, working with DnaA (which initially recognizes and melts the origin, oriC) and single-stranded binding proteins (SSB) that stabilize the separated strands and prevent reannealing. Topoisomerases (e.g., topoisomerase II/gyrase) work ahead of helicase to relieve the torsional strain and supercoiling generated by unwinding. Clinically, defective helicase function is implicated in several genetic diseases: mutations in RECQ-family helicases cause Bloom syndrome (BLM gene), Werner syndrome (WRN gene), and Rothmund-Thomson syndrome, all associated with genomic instability, premature aging features, and increased cancer risk. Helicase is also a target in antiviral therapy (e.g., helicase-primase inhibitors like pritelivir for HSV) and is essential to DNA replication highlighted in cell cycle and molecular biology topics for boards.
